What Is Leucovorin?
Leucovorin is a form of folate often used in medicine to support patients after chemotherapy. In recent years, researchers have studied its role in autism therapy. Early findings suggest that leucovorin may enhance speech and communication in children with autism, particularly those with specific challenges in folate metabolism.
How Folate and Autism Are Connected
Folate (vitamin B9) is essential for brain development. But many children with ASD have cerebral folate deficiency, often linked to antibodies that block folate receptors in the brain. These folate receptor autoantibodies (FRAA) are present in up to 70% of children with autism, compared to only 5–10% of the general population.
When folate cannot enter the brain, speech, language, and learning are affected.
How Leucovorin Helps
Leucovorin (folinic acid) bypasses the blocked folate receptors by using an alternative pathway, restoring folate levels in the brain. Unlike regular folic acid supplements, high-dose leucovorin can cross the blood-brain barrier and support healthy brain function.

Improved social interaction
Better developmental progress
Typical dosing ranges from 0.5 to 2 mg per kg of body weight daily, up to 50 mg/day, always under medical supervision.
Safety and Side Effects
Leucovorin is generally safe for children. Some individuals may experience increased energy or restlessness during the first few weeks, which typically resolves within 6 to 9 weeks. Gradual dosing helps reduce these effects. No significant concerns have been reported in lab tests for blood, liver, or kidney function.
